Despite recent improvements in treatment, cancer remains a huge public health burden. Cytotoxic treatments play a critical role in modern cancer therapy. Unfortunately, traditional cytotoxic agents are associated with significant toxicity. In an effort to discover new oncology drugs with improved clinical benefits and fewer side effects, drug developers have committed substantial resources to identifying novel cancer targets and compounds that interact with those targets. These efforts have been facilitated by a plethora of technologies developed in recent years to generate and exploit genomic and proteomic information. In this report, we review novel cancer drug targets and the drugs under development directed at these targets.
