Glioblastoma multiforme (GBM) is a highly aggressive form of glioma, a cancer arising from astrocytes, and is the most common type of brain tumor in adults (Kanderi et al., 2024). The distinction of "multiforme" comes from the wide range of genetic expressions characteristic of the cancer, complicating treatment (Kanderi et al., 2024). Glioblastoma multiforme is currently incurable, with a median survival time of 15 months and a 7.2% five-year survival rate (Kanderi et al., 2024). Gold standard treatment is a combination of surgical resection, followed by the "Stupp protocol" comprising radiotherapy and temozolomide (Kanderi et al., 2024).

In 2021 the WHO differentiated between two types of glioblastomas - isocitrate dehydrogenase (IDH)-wild type and IDH-mutant. The new classification listed IDHWT as glioblastoma and re-labeled IDH-mutant as diffuse astrocytoma grade IV (Stoyanov et al., 2022). For the purposes of this model, both IDH-WT and IDHmutant forms are classified as GBM, though the differentiation is noted in relevant segmentations.

Scope

  • The model includes a 10-year epidemiological forecast for the diagnosed incident cases and five-year diagnosed prevalent cases of GBM. Diagnosed incident cases are segmented by age (starting from 0 up to 85 years and older by 5-year age groups) and sex. Additionally, the diagnosed incident cases of GBM are segmented by origin (primary or secondary), biomarker (MGMT methylated/unmethylated, EGFR VIII positive, EGFR amplification positive), and mutation (IHD1/IHD2 mutation, TERT promoter mutation, BRAF-V600E mutation, HLA-A2 mutation, and NTRK mutation).
  • The model also includes includes a forecast of diagnosed incident cases of central nervous system (CNS) cancer by subtype (glioblastoma, meningioma, ependymoma, oligodendroglioma, CNS lymphoma, and other).
