Wilson Disease Emerging Therapy and TPP Insights

Thelansis's "Wilson Disease Emerging Therapy, with Unmet Needs and TPP Insights Report - 2026" provides a comprehensive analysis of the emerging competitive landscape, unmet needs, target product profiles (TPPs), trial designs, and KOL insights on key emerging therapies and key drug development opportunities in the indication.

Wilson Disease Overview

Wilson disease is an autosomal recessive disorder caused by pathogenic variants in ATP7B, which encodes a copper-transporting ATPase required for biliary copper excretion. Impaired ATP7B function causes copper accumulation in the liver and, with progression, the brain, cornea, and other tissues. Clinical manifestations vary and may include hepatitis, cirrhosis, acute liver failure, tremor, dystonia, dysarthria, movement disorders, psychiatric symptoms, and Kayser-Fleischer rings. Treatment aims to remove excess copper and prevent further accumulation using penicillamine, trientine, and/or zinc. Liver transplantation is reserved for selected patients with acute liver failure or advanced liver disease. Despite effective therapies, lifelong treatment, adherence demands and persistent neurological symptoms remain challenges. These limitations are driving development of disease-modifying strategies, including ATP7B gene replacement and gene-editing approaches such as UX701 and PM577a.

Key Highlights

  • In the United States, diagnosed Wilson disease cases are estimated at 7,574 in 2026.
  • Wilson disease is a rare inherited disorder that causes copper accumulation in the liver, brain, and other organs.
  • Delayed diagnosis can lead to serious hepatic and neurological complications.
  • Unmet need remains for treatments that improve copper control and support long-term organ health.

Market Overview

  • The Italy Wilson disease market is projected to grow from $25M in 2026 to $33M by 2036.
  • Market growth is driven by:
  • Continued use of copper-lowering therapies and improved disease management
  • Advances in diagnosis and development of novel treatment approaches
  • Future growth will depend on therapies that offer effective long-term copper control, improved tolerability, and better patient outcomes.
